A good method for obtaining skin, muscle, bone (and posterior kidney in the case of fish with a thin retroperitoneal kidney) is to take a thin cross section through the entire body. Sample a 1/2-inch wedge of skin and muscle from the cross section and preserve the dark posterior kidney tissue as well.
You may want to sample part of the backbone and preserve it in Bouin's fixative, which will have better penetration and soften the bone tissue for histological processing. |
